木瓜凝乳蛋白酶对长春新碱杀伤鼠肝癌细胞hepa-6的促进作用

摘要 目的 测定木瓜凝乳蛋白酶与长春新碱 (VCR)联合使用对鼠肝癌细胞hepa - 6的细胞毒性作用。方法 实验分为 3组 :酶组、VCR组、酶 +VCR组 ,在培养不同时间 ,3种不同浓度的酶与VCR单独或联合使用时 ,采用MTT法检测对鼠肝癌细胞hepa - 6的细胞毒性。同时分析培养时间对木瓜凝乳蛋白酶活力的影响。结果 木瓜凝乳蛋白酶对hepa - 6细胞的半抑制浓度 (IC50 )约为 1 2 0 0 μg/ml,随浓度的增加对细胞的抑制率增加。 3种不同浓度的酶 (5 0 ,1 0 ,2 μg/ml)与VCR(1 0 μg/ml)联合使用时 ,在细胞培养至 4 8h前 ,木瓜凝乳蛋白酶均能促进VCR杀伤鼠肝癌细胞hepa - 6 ;这个现象与酶在生理盐水中的活力变化是一致的。结论 木瓜凝乳蛋白酶对鼠肝癌细胞hepa - 6具有细胞毒作用 ,对长春新碱杀伤鼠肝癌细胞hepa - ObjectiveTo study the cytotoxicity of chymopapain and vincristine(VCR) on mouse hepatoma cell line hepa-6 cultured in vitro. MethodsThree groups were divided: chymopapain group, VCR group and chymopapain-VCR group. The cytotoxicities were measured by MTT method when Hepa-6 cell were treated by three different concentration of chymopapain and VCR alone or in combination with different culture time. The activity of chymopapain was measured in different culture time. ResultsChymopapain inhibited the growth of mouse hepatoma cell line Hepa-6(IC 50 was about 1?200 μg/ml ) in a dose-dependent manner. Synergistic effect was observed when three different concentration chymopapain(50,10 and 2 μg/ml ) was in combination with VCR(1 0 μg/ml ) for 48 hour, in accordance with the change of enzyme activity of chymopapain. ConclusionChymopapain has cytotoxicity on hepa-6 cultured in vitro and has synergistic effect when combined with VCR.
