摘要
目的 利用生物信息学方法分析 DIF14 / L MBR1蛋白的分子进化情况并分析其同源蛋白的保守区。方法 利用BL AST同源比较分析寻找 Gen Bank中与 DIF14 / L MBR1高度同源的蛋白序列并下载 ,应用 Clustal X软件对蛋白序列进行多序列对齐 ,绘制进化树 ,利用 SAM- T99软件分析多个序列之间的保守区。 TMpred软件对下载的各种同源的蛋白序列进行跨膜区预测。结果 DIF14 / L MBR1蛋白在各种真核生物中存在高度保守的同源蛋白 ,大部分蛋白属于九次跨膜蛋白 ,各蛋白之间的氨基酸一致性与生物亲缘关系的远近有密切关系 ,人 DIF14 / L MBR1与lipocalin- interacting membrane receptor基因可能有相同的祖先基因 ,但两者功能有所不同。结论 DIF14 / L MBR1蛋白是真核生物中高度保守的基因 ,属于九次跨膜蛋白 ,但在细胞内具体执行的生物学功能有待进一步研究。
Objectives To analyze the molecular evolution of DIF14/LMBR1 protein and the conserve domain of the homologues by using bioinformatics. Methods Proteins homolog to DIF14/LMBR1 were searched in GenBank by using BLAST(Basic Local Alignment Search Tool) and the amino acid sequences were downloaded. The amino acid sequences were aligned and the phylogenetic tree were constructed by using the Clustal X software. The conserve domain were analyzed between the multiple sequences by using SAM-T99, Analysis of the transmembrane region of the multiple amino acid sequences were doing by using TMpred software. Results There are high conserved proteins homolog to DIF14/LMBR1 in different eukaryotic species. Most of the proteins belong to nine trans-membrane protein. There maybe same ancestor gene of DIF14/LMBR1 and lipocalin-interacting membrane receptor.Conclusion DIF14/LMBR1 is highly conserved gene of eukaryotic organisms,the amino acid identity of DIF14/LMBR1 between human and different species were related to the taxonomy distance.The function of DIF14/LMBR1 need further research.
