摘要
目的 探讨多氯联苯同系物PCB1 53 对胚胎生长发育的影响及其作用机制。方法 于体外将孕 8.5d小鼠胚胎置入不同浓度PCB1 53 ,应用植入后全胚胎培养技术研究PCB1 53 对胚胎生长发育的影响 ;透射电镜观察PCB1 53 对卵黄囊的损害。结果 PCB1 53 对胚胎发育具有明显影响且呈剂量 效应关系。 0 .1 0ng/ml组除卵黄囊循环稍差外 ,其余指标与对照组一致或差异不显著 (P>0 .0 5 ) ;1 .0 0ng/ml组明显影响卵黄囊循环、头长、颅臀长、体节数、中脑、后脑、翻转、鳃弓和上下颌突等指标 ,与对照组比较P <0 .0 5 ;1 0 .0 0ng/ml组 ,各指标进一步降低 (P <0 .0 1 ) ,胚胎致畸率升高并出现死亡 ;电镜发现 ,PCB1 53 处理的各实验组卵黄囊超微结构遭到不同程度破坏。结论 PCB1 53 使胚胎发育迟缓、具有发育毒性和致畸性 ,作用的敏感部位主要是卵黄囊、头长、颅臀长、中脑和鳃弓 ;卵黄囊的超微结构损害与PCB1 53 致胚胎发育异常有关。
Objective To explore the effects of polychlorinated biphenyl congener 153(PCB 153 ) during pregnancy on embryonic development and its mechanism.Methods Mouse embryos of gestational day 8.5 were exposed to different doses of PCB 153 in vitro.Post implantation whole embryo culture (WEC) technique and transmission electron microscopy (TEM) were used to investigate the effects of PCB 153 on embryonic development and ultrastructures of visceral yolk sac (VYS) cells.Results PCB 153 had obvious effects on the development and growth of embryos with a dose response relationship.0.10ng/ml group caused no significant differences to embryonic development and organ morphological scores except for slight impairment of VYS circle,as compared with the non PCB 153 treated controls ( P >0.05).1.00ng/ml group led to the significant decrease of scores of VYS circle,head length,crown rump length,number of somites,midbrain,hindbrain,trunk turning,branchial arch,maxillary process and mandibular processs ( P <0.05);10.00ng/ml group resulted in progressive reduction of scores of indexes ( P <0.01),increased incidence of embryo malformation and fetal death;ultrastructures of VYS cells were damaged diversely in all PCB 153 treated experimental groups.Conclusions PCB 153 causes fetal growth retardation,developmental toxicity and teratogenesis.VYS,head length,crown rump length,midbrain and branchial arch are the targets of PCB 153 effects.Its effects on embryonic development are related with the damage of ultrastructures of VYS cells.
