摘要
目的 观察不同剂量胰岛素样生长因子 1(1GF 1)对海水浸泡的大鼠肺泡Ⅱ型上皮细胞(AT Ⅱ )的抗凋亡作用。方法 建立体外培养的大鼠AT Ⅱ细胞海水浸泡模型 ,加入不同剂量的胰岛素样生长因子 1,采用流式细胞仪 (FCM)检测AT Ⅱ细胞凋亡和坏死率。以 2 ,3 苯基溴化四唑 (MTT)染色记数法绘制活细胞生长曲线。结果 海水可直接导致AT Ⅱ细胞损伤 ,表现为AT Ⅱ细胞坏死和凋亡发生率增高 ;IGF 1能促进AT Ⅱ细胞增殖并降低其凋亡率 ,在实验剂量范围内 ,其抗凋亡和促增殖作用呈剂量依赖性增强。结论 IGF 1对海水所致的AT Ⅱ细胞凋亡有直接的抑制作用。
Objective To study the effects of insulin-like growth factor-1 (IGF-1) on seawater induced apoptosis of cultured adult rat alveolar type II cells(AT-Ⅱ).Methods Cultured AT- II cells were exposed continuously to sea water for 24 h.Apoptosis and necrosis rates of AT- Ⅱ cells were detected by flow cytomer (FCM). The proliferation curve was drawn.Results The injuries which included necrosis and apoptosis of cultured AT- Ⅱ cells could be stedily induced by sea water steady in this model.IGF-1 might not only decrease the apoptosis ratio but also promote the proliferation of AT-Ⅱ cells. It showed a dose-dependent effect of IGF-1 on AT-Ⅱ cells.Conclusion IGF-1 can directly inhibit the apoptosis induced by sea water in cultured AT-Ⅱ cells.
