摘要
检测血液凝固性和血小板免疫粘附功能是研究血瘀证实质和活血化瘀药理作用的重要手段。为了探讨雪莲活血化瘀的药效及作用机制,本实验对沙门氏菌性“热毒血瘀”模型兔给药前后的血小板数、血小板电泳速率、血小板聚集性、体外血栓形成、血浆纤维蛋白原含量、优球蛋白溶解时间及血小板免疫粘附功能进行了检测。证明雪莲对二磷酸腺苷(ADP)诱导的正常兔和“热毒血瘀”模型元的血小板聚集性具有很强的抑制作用,其抑制率分别为21.1±2.9%(±Sx)和25.6±2.7%(±Sx),且对模型兔血小板聚集性的抑制作用要比对正常兔的强(P<0.01);雪莲对亢进的血小板免疫粘附功能具有抑制作用。此外,雪莲还具有抗体外血栓形成及抗机体纤溶活性降低的作用。但雪莲对血小板的表面电荷无影响,对血小板的生成也无明显促进作用。结果表明,雪莲的活血化瘀作用机制与其促进机体纤溶活性和降低血液凝固性有着密切关系。
Saussurea Gossypiphora is a traditional chinese medicine comm only used by the Tibetan veterinary surgeon for the purpose of promoting the blood circulation, regulationg the menstruation and reinforcing the deficient, etc. In order to study the mechanism of activating blood circulation and removing stasis of Sgossypiphora. The method such as platelet count, determination of platelet electrophoresis rate, platelets aggregation test, extracorporeal thrombosis test, determination of fibrinogen content in plasma and euglobulinlysis time as well asplatelets immune adherence determined by saecharomycete agglutination technique in vitro were employed to investigate it's effect on blood coagulation and platelets immune adherence in blood stasis model of rabbit caused by febrile-toxicution. the results showed that the Saussurea Gossypiphora not only could markedly inhibit the agglutination of platelet induced by ADP(adenosine diphosphate) but also the inhibion to the agglutination of platelets from blood stasis models was higher than that of platelets from normal rabbit(inhibition rate were 25.6±2.7 and 21.1±2.9 respectively). Saussurea Gossypiphora could inhibit the hyperfunction of platelets immune adherence and extracorporeal thrombosis, and could promote the activity of fibrinolysis. But it could not influence the decrese of surface chargedensity of platelets and the amount of platelets in blood stasis model caused by febriletoxication. on the basis of these results, the author conclude that the association of mechanism of activating blood circulation and removing stasis of S. Gossypiphora is colsely with promoting fibrinolysis and reducing blood coagulation.
