摘要
目的 :探讨哮喘患者的外周血单核细胞 (peripheralbloodmonocyte,PBMC)来源的树突状细胞 (dendriticcell,DC)表面协同刺激分子CD1a、CD83、CD4 0、CD86的表达 ,以及其激发同种异体T淋巴细胞的能力 ,为进一步研究哮喘的发病机制及治疗提供新的思路。方法 :取哮喘患者及正常人外周血以Thomas法分离出PBMC ,然后加入相关的细胞因子及抗原培养并诱导出相应的成熟的DC细胞。用流式细胞仪 (fluorescentactivatedcellsortor,FACS)检测DC表面协同刺激分子CD1a、CD83、CD4 0、CD86的表达。成熟的DC与同种异体T淋巴细胞反应后用细胞计数板计数得出T淋巴细胞总数。结果 :哮喘患者DC的CD86表达比正常人高 (t =7.35 ,P <0 .0 0 1 ) ,但其激发同种异体T淋巴细胞的能力较对照组无明显增强。结论 :哮喘患者可表达较高水平的CD86并且有较强的激发的能力 。
Objective: To study the expression of the phenotype CD1a、CD83、CD40、CD86 on monocyte derived dendritic cell(DC) in atopic asthmatic patient,and their potentials to stimulate the proliferation of allogenic T cell in healthy human. Methods: Thomas method was adopted.Peripheral blood monocytes (PBMC s) were isolated from asthmatic patients and healthy human peripheral blood.Then put PBMC in medium with rhGM CSF, and rhIL 4 and LPS were used to induce the maturity of DC .The expression of the CD1a,CD83,CD40,CD86 on mature DCs were examined by fluorescent activated cell sortor (FACS),and their potentials to stimulate the proliferation reaction of allogenic T cells from healthy human were observed. Results: The results showed that asthmatic patient had increased expression of the CD86 on DCs ( t =7.35, P< 0.001 ). At the same time, the asthmatic patients had increased the ability of DCs inductive allogenic T cells reaction compared with the normal control subjects. Conclusion: The expression levels of CD86 are higher in asthmatic patient than in the healthy control group.This result suggests DCs may play an important role in asthma.
