苦参碱对心肌成纤维细胞细胞周期的影响及其机制 被引量:24

摘要 目的 :观察苦参碱 (Mat)对血管紧张素Ⅱ (angiotensin ,AngⅡ )诱导的心肌成纤维细胞细胞 (CFs)增殖的影响并探讨其机制。方法 :体外培养新生Wistar大鼠的心肌成纤维细胞 ,四氮唑盐 (MTT)比色法检测细胞增殖 ,流式细胞分析仪测定细胞周期及细胞周期蛋白表达。结果 :①随苦参碱浓度的增加 ,CFsMTT值呈递减趋势 ,且均明显低于AngⅡ组 ;②苦参碱可使CFs的S期细胞百分率下降 ,G0 /G1 期百分率上升 ,与AngⅡ组比较差异较显著(P <0 .0 5 ) ;③AngⅡ可使CFsP2 7表达阳性率降低 ,而苦参碱可促进CFsP2 7表达。结论 :苦参碱可抑制AngⅡ诱导的心肌成纤维细胞增殖 ,P2 Objective: To study the effects and mechanisms of matrine on the proliferation induced by AngⅡ.Methods: Cultured cardiac fibro blasts (CFs) of neonatal Wistar rats were treated with matrine. CFs prolifertion was measured by thiazolyl blue(MTT) assay. Cell cyclin distribution and P27 expression were determined with flow cytometre.Results: ①Ang Ⅱ increased value of CFs in concentration dependent manner .The value of matrine treated group was significantly lower than that of the AngⅡgroup. ②The percentage of CFs in S stage was decreased significantly by matrine, while with a significant increase in the percentage of G 0/G 1 stage cells.③P27 expression in CFs was downregulated by AngⅡ,which was signficantly higher than that of the control.Matrine could increase the expression of P27 in CFs.Conclusion: Matrine inhibits effectively the prolifertion of cultured rat CFs induced by angiotensinⅡ. P27 may be a modulating factor in CFS proliferation.
