摘要
为了从离体细胞水平探讨昆虫对苏云金芽孢杆菌杀虫晶体蛋白的部分抗性机制,本文采用活化的Cry1Ac 毒素对粉纹夜蛾BTI-TN-581-4细胞连续筛选86代,获得了高水平抗性细胞,研究了其某些特性。它对Cry1c 产生了低水平的交互抗性,对低渗溶液的耐受性显著增强,双向电泳图谱表明抗性细胞膜蛋白组分发生了明显的变化。膜蛋白组分的变化可能导致了筛选细胞的耐低渗透压和抗Cry1C。
The cell line (BTI-TN-5B1-4) of Trichoplusia ni had been selected by Bacilllusthuringiensis (Bt) toxin Cry1Ac for 86 generations continuously.A high level of resistance wasachieved and the partial characterizations of the resistance-selected BTI-TN-5B1-4 cells werestudied in order to explore the mechanism of the resistance to Bt toxin.The cells selected byCry1Ac had low level of cross-resistance (19.7 fold) to Cry1C,their ability of tolerance in thehypotonic solution increased evidently (2.5 folds),and there were differential membrane proteinsbetween resistance-selected and unselected susceptible cells on two dimensional eleetrophoresismaps.We supposed the changes of membrane proteins in resistant cells would be linked to thecross-resistance to Cry1C and the increase of tolerance in hypotonic solution.
