Culture model in vitro of fetel arterial smooth muscle cells and their growth characters

摘要 目的 探讨胎儿动脉导管、主动脉及肺动脉平滑肌细胞体外培养方法 ,明确不同平滑肌细胞的原代和传代后生长情况。方法 采用改良贴壁法 -玻片压迫组织贴壁方法进行胎儿动脉导管、主动脉及肺动脉平滑肌细胞的原代培养 ,并将细胞进行传代后培养 ,同时对各细胞的生长进行观察。结果 ①采用改良贴壁法可使原代细胞良好生长 ,动脉导管的平滑肌细胞无论是从组织块游出时间还是生长呈“峰 -谷”样时间和细胞首次传代时间都比肺动脉和主动脉的要长 ;②在细胞的继代培养期 ,动脉导管平滑肌的生长周期长于主动脉 ,主动脉的平滑肌生长周期长于肺动脉。结论 动脉导管、主动脉及肺动脉平滑肌细胞体外生长存在差异。 Objective To investigate culture method in vitro of fetel smooth muscle cells from aorta, pulmonary artery and arterial duct and determine growth characters of different cells in primary state and passage state.Methods Fetel smooth muscle cells from aorta, pulmonary artery and arterial duct were primarily cultured by modified explant-attached method that tissues were pressed with glass slice and all cells were continuously cultured after passage, at same time, their growth were observed.Results ①Primary cells from aorta, pulmonary artery and arterial duct grew very well. Cells from fetel arterial duct spent more time than ones from aorta and pulmonary artery on no matter their leaving tissue or growth forming “hills and valleys” or first passage; ②In passage culture, growth cycle of cells from fetel arterial duct was longer than that from aorta and growth cycle of cells from was longer than that from pulmonary artery.Conclusion There were differences among growths of fetel smooth muscle cells in vitro from aorta, pulmonary artery and arterial duct in vitro.
