外源DNA导入小麦变异后代的抗旱耐盐性初步鉴定及RAPD验证 被引量:7

摘要 通过对导入外源DNA的小麦变异后代进行抗旱性和耐盐性的鉴定 ,筛选到四个抗旱耐盐性较好的小麦新品系。引物S317的RAPD验证结果表明 ,四个变异后代均有两条异于受体的特异谱带 ,其分子量分别为 6 5 0bp、6 80bp。其中分子量为 6 5 0bp的谱带在四个变异后代和供体中均具有 ,而受体却没有。这可能是玉米基因组中的DNA片段整合到受体的基因组中 ,导致受体中与抗性有关的基因结构发生了改变 ,从而提高了受体的抗旱耐盐能力。室内考种结果表明 。 By the test of salt and drought resistance of wheat variation offspring produced by exogenous DNA introduction, four variation offspring strains were screened with better salt and drought resistance. RAPD result with the primer S317 indicates that four variation offspring strains all have two exceptionally different belts from the receptor, and molecular weights are respectively for 650bp and 680bp.Four variation offspring strains and the donor have the exceptionally different belts which molecular weights is 650bp, but not the receptors. This probably is DNA's part conformity in the maize gene group to the gene group of reception body,and the gene structure that leads to resistance has changed, thus improved reception body.It endures salt and drought resistance.The part of agronomy properties of four wheat variation strains studied indoors are superior to that of the reception bodies.
