绵羊卵巢腔前卵泡的分离与体外培养

摘要 分离方法和绵羊年龄对绵羊卵巢腔前卵泡的分离效果有显著影响。从每只羊获得腔前卵泡的总数看,机械分离法(89.5)和酶消化法(103.3)差异不显著,而正常卵泡数和正常卵泡率机械分离法(27.9,31.2%)明显高于酶消化法(19.1,18.5%);采用机械分离法,每只初情期前绵羊获得的腔前卵泡总数和正常卵泡数(125.6,35.1)均高于成年绵羊(60.4,16.1),而正常卵泡率(28.1%比26.8%)差异不显著。腔前卵泡在体外培养6 d,培养液中添加20ng/mL的IGF-I,培养结束时卵母细胞直径增幅为34.8 μm,与对照组的29.9 μm差异不显著。 Isolation method and sheep age had significant effects on the isolation efficiency of sheep preantral follicles.Per sheep mechnical isolation obtained the same total preantral follicles(89.5) with that of enezmy digestion (103.3)(P>0.05) per sheep,but normal follicle rate(31.2%) of mechnical isolation was high than that (18.5%) of enezmy digestion (P< 0.05).By mechnical isolation, per preputuberty sheep obtained higher total preantral follicles and normal preantral follicles(125.6,35.1)than that of adult sheep (60.4,16.1)(P<0.05), but normal follicle rate did not differ(28.1 % vers 26.8% ) .Preantral follicles were cultured in vitro for 6 days, no difference was observed in oocyte growth (34.8 μm vs 29.9 μm) between IGF- I (20 ng/mL) treated follicles and controls.
