摘要
应用体外基因扩增技术放大宫颈癌组织中H-ras癌基因的部分DNA顺序,再以特异突变的寡核苷酸探针杂交检测宫颈癌中H-ras癌基因第12位密码子的G→T突变,结果在33例宫颈癌中发现有10例存在这种突变,而17例慢性宫颈炎无1例发生突变。提示:H-ras癌基因第12位密码子G→T突变可能是宫颈癌中癌基因激活的方式之一。
The Polymerass chain reaction (PCR) was used to amplify the DNAsequence of H-ras oncegene in the tissue of cervical cancer, and the G- Tmutartion at . code 12 of the H-ras oncogen was detected by hybridization whth mutation specific oligon-ucleotide probes. The result showed that the G-T mutation rate was 30.3%(10/33) in human cervical cancers, but absent in 17 chronic cervicitis. This result implicated that G-T mutation at codon 12 of the. H-ras oncogene may be one of the oncogene activated mechanisms in cervical cancers.
