摘要
本文报道复方丹参、地龙、野木瓜注射液对体外培养的人鼻咽癌细胞株CNE-2的放射增敏作用。在96孔塑料微量细胞培养板中培养,均接受2 Gyγ射线照射,然后继续培养,在倒置显微镜下观察记录每孔克隆数,将用药组与单纯放射组比较。方差分析表明各加药组均有明显的放射增敏作用,其增敏率27.1%~54.3%(P<0.001)。用美兰比色法测定细胞氧耗结果表明,复方丹参、地龙、野木瓜都可明显促进癌细胞氧耗增加。
The radiosensitization activity of Chinese herbal medicines Injectio Sal- via mitltiorrhiza complex, Pheretima asiatica, and Stauntonia chinensis on cultured human na-sophary ngeal carcinooma cell line (CNE-2) in vitro was studied. The CNE-2 cells were seeded into 96 well microcultured plate, 50 cells per well, 24 wells as one group with or without medicine in media, cultured for 12-15 hrs. Then the cells were irradiated by 60Co in a obse of 2 Gy y-ray and continuously cultured for 70-72 hrs. The cell clones(≥8 cells) per well were observed and counted by inverted microscope. The inhibition rate of colony formation in the irradiated combined with medicine group increased by 27.05%-54. 27% over the irradiated alone group. The differences were statistically highly significant(P<0.0001). Using methylene blue colorimetric method to assay the O2 consumption, the results showed that all of the three Chinese herbal medicines can promote apparently O2 consumption of CNE-2 cells.
