摘要
目的探讨银屑病皮损区角质形成细胞p16基因D9S171,D9S1604微卫星位点杂合性缺失(lossofheterozygosity,LOH)与银屑病发病的关系。方法采用聚合酶链反应-变性聚丙烯酰胺凝胶电泳-银染技术对体外原代培养的银屑病皮损区和非皮损区角质形成细胞LOH进行分析。结果23例银屑病患者皮损区体外培养的角质形成细胞在D9S171和D9S1604两个微卫星位点,LOH分别为5例和10例。在非皮损区的相应位点LOH分别为2例和3例。D9S1604LOH频率明显高于非皮损区角质形成细胞(P<0.05);寻常性银屑病患者皮损区角质形成细胞中两位点LOH发生频率分别为23.5%(4/17)和47.1%(8/17),非寻常性银屑病分别为16.7%(1/6)和33.3%(2/6),两型银屑病两位点LOH发生频率相比差异均无显著性(P>0.05)。结论D9S1604微卫星位点LOH与银屑病的发生发展相关,而与临床类型相关无显著性。
Objectives To detect the loss of heterozygosity(LOH)frequencies of microsatellite loci D9S171and D9S1604in p16gene of psoriatic keratinocytes,and to study the correlation between mi-crosatellite LOH of p16gene and the development of psoriasis.Methods By the use of polymerase chain reaction(PCR)-denaturing polyacrylamide gel electrophoresis-silver staining,LOH was detected with23sam-ples of keratinocytes from psoriatic lesions and non-lesion skin.Results LOH was identified at loci D9S171and D9S1604in5and10out of23keratinocyte samples from LOH-exhibited psoriatic lesions,and in2and3of keratinocyte samples from psoriatic non-lesion skin,respectively.The frequency of LOH at D9S1604was significantly higher in psoriatic lesion samples than that in psoriatic non-lesion skin(P<0.05).The frequencies of LOH at D9S171and D9S1604were23.5%(4/17)and47.1%(8/17)in ker-atinocyte samples from lesions of psoriasis vulgaris,and16.8%(1/6)and33.3%(2/6)in those from other clinical types of psoriasis,respectively.There was no significant difference of LOH frequencies between sam-ples taken from psoriasis vulgaris and other types of psoriasis.Conclusions The frequencies of LOH at the microsatellite locus D9S1604in p16gene are associated with the development of psoriasis.There is no sig-nificant correlation between LOH frequency and clinical types of psoriasis.
