摘要 目的 研究大鼠成骨细胞与Bio-oss骨粉体外复合培养对成骨细胞成骨效能的影响,探讨Bio-oss作为骨组织工程支架材料的组织相容性。方法 采用原代骨髓基质干细胞(MSCs)培养技术及定向诱导培养技术获得大鼠成骨细胞,分为实验组与对照组。实验组细胞与Bio-oss骨粉复合培养,对照组细胞单独培养。比较两组成骨细胞形态学特点、碱性磷酸酶(ALP)活性、贴壁率、增殖活力及蛋白含量。结果 成骨细胞与Bio-oss骨粉复合培养后,细胞形态、增殖活力、成骨效能(ALP活性、蛋白含量)与单纯成骨细胞培养无明显差异。结论 Bio-oss骨粉能满意模拟天然骨支架,可作为骨组织工程中较理想的支架材料。 Objective To investigate the osteogenesis of osteoblasts of SD rats cultured with Bio-oss powder in vitro and study the biocompatibility of the Bio-oss powder as a scaffolds in tissue engineering with the osteoblasts. Methods Osteoblasts of SD rats were cultured and divided into 2 groups. Experiment osteoblasts were seeded onto Bio-oss powder and the cell/Bio-oss composites were cultured in vitro. Control osteoblasts were cultured alone. Then cell adhension, proliferation, ALP activity and protein concentration were measured. Results There were no significant difference in activity of ALP, proliferation, cell adhension and protein concentration between osteoblasts cultured with Bio-oss and those cultured alone. Conclusion It was suggested that Bio-oss has great cellur biocompatibility. It is an ideal scaffold material for bone tissue engineering.
