摘要 目的 检测PTEN、p2 7kip1、cyclinE 3种蛋白在非小细胞肺癌组织的表达及与临床病理特征的关系。 方法 在 6 0例非小细胞肺癌原发灶 (包括 37例鳞癌和 2 3例腺癌 )中应用免疫组织化学S P法检测 3种蛋白的表达。结果  6 0例肺癌原发灶中PTEN、p2 7kip1、cyclinE 3种蛋白表达阳性率分别为 :4 8.3% ;4 1.7% ;73.3%。PTEN与淋巴结转移显著相关 (P <0 .0 5 ) ,p2 7kip1、cyclinE与淋巴结转移无关 (P >0 .0 5 ) ;这 3种蛋白表达均与肿瘤细胞分化程度显著相关 (P <0 .0 5 ) ,与组织学类型无关 (P >0 .0 5 )。 6 0例非小细胞肺癌组织中 ,PTEN的表达与 p2 7kip1呈显著正相关 (P <0 .0 1) ,而与cyclinE无明显相关 (P >0 .0 5 ) ;p2 7kip1与cyclinE的表达之间呈显著负相关 (P <0 .0 1)。 结论 PTEN明显地抑制了非小细胞肺癌的浸润和转移 ,p2 7kip1表达缺失与肺癌细胞的分化有关 ,PTEN/p2 7kip1衰老诱导途径在非小细胞肺癌的恶性进展中起着很重要的作用。 Objective The relationships between the expressions of PTEN, p27 kip1 , cyclin E proteins and the clinicopathological characteristic of NSCLC were investigated. Methods Immunohistochemical methods were applied to detect the expression of PTEN, p27 kip1 , cyclin E in 60 surgical specimens form NSCLC patients (37 squamous cell carcinomas and 23 adenocarcinomas). Results In 60 NSCLC, the positive rates of PTEN, p27 kip1 and cyclin E proteins were 48.3%, 41.7% and 73.3%, respectively.The positive expression of PTEN protein was highly associated with lymph node metastasis( P <0.05), but not the p27 kip1 and cyclin E expressions( P >0.05). Positive rates of these three proteins were closely associated with the cellular differentiation ( P <0.05), but not with the pathological types( P >0.05). PTEN expression was positively associated with p27 kip1 expression( P <0.01),but not with cyclin E expression ( P >0.05). An inverse correlation was present between cyclin E and p27 kip1 expressions( P <0.01). Conclusion PTEN obviously inhibits invasion and metastasis of NSCLC; loss expression of p27 kip1 is correlated with lung cancer cell differentiation, PTEN/p27 kip1 senescence inducing pathway may play an important role in NSCLC malignant progression.
