摘要 目的 探讨单纯疱疹病毒胸苷激酶自杀基因 (HSV TK) /丙氧鸟苷 (GCV)系统和化疗联合应用提高舌鳞癌治疗疗效的可能性。方法 舌鳞癌Tca8113细胞接种于裸鼠右侧腋下 ,随机分成 8组进行治疗 ,观察各组移植瘤的生长状态及组织病理学改变 ;计算抑瘤率和群体倍增时间 ;析因分析化疗药物与基因治疗的相互作用。结果 与对照组比较 ,各治疗组移植瘤的生长均明显受到抑制 (P <0 .0 0 1) ,移植瘤的群体倍增时间延长 ;双因素析因分析显示 5 FU与HSV TK/GCV之间、DDP与HSV TK/GCV之间 ,以及DDP +5 FU与HSV TK/GCV之间均存在协同的抑瘤作用 (P <0 .0 0 1)。光镜下各治疗组移植瘤组织内均可见囊性变和大片的灶性坏死。结论 HSV TK/GCV基因治疗与 5 FU和 /或DDP联合应用可提高对舌鳞癌的治疗疗效。 Objective To investigate the therapeutic efficiency of combination of herpes simplex virus thymidine kinase (HSV TK)/ganciclovir(GCV)system with 5 fluorouracil (5 FU) and/or cisplatin (DDP) in the treatment of tongue squamous cell carcinoma. Methods Tca8113 cells were inoculated subcutanously in the right flank of BALB/c nude mice. Mice were randomly assigned to 8 groups and treated. The tumor growth and the histopathological changes were observed and the inhibition rate and tumor doubling time were calculated. The anticancer effects between 5 FU/DDP and HSV TK/GCV were analyzed by factorial experiment. Results Compared with control group, mice in treated groups (group ② ⑧) demonstrated significant tumor regression ( P <0.001) and the tumor doubling time prolonged. Cooperative anticancer effects were demonstrated between 5 FU and HSV TK/GCV, DDP and HSV TK/GCV,DDP+5 FU and HSV TK/GCV by factorial experiment. Cyst formation and substantial tumor necrotic areas in each treated group were observed under light microscope. Conclusion The cooperative anticancer effect between chemotherapy (5 FU±DDP) and HSV TK/GCV system can be found in the treated of tongue squamous carcinoma.
