超声辐照对血管紧张素Ⅱ诱导的牛主动脉平滑肌细胞增殖的影响 被引量:1

摘要 为观察不同剂量的超声辐照对牛主动脉平滑肌细胞 (CASMC)增殖的影响 ,寻找出抑制细胞增殖的最佳辐照剂量 ,利用体外培养的牛主动脉平滑肌细胞 ,用细胞计数法、MTT法、3H 胸腺嘧啶核苷 (3H thymidine,3H TdR)掺入法测定细胞增殖率的变化 ,观察不同浓度的血管紧张素Ⅱ(AngⅡ)对血管平滑肌细胞增殖的影响以及不同剂量的超声辐照对AngⅡ诱导的血管平滑肌细胞增值的预防和逆转作用。细胞计数、MTT法、3H TdR掺入法均显示 1 0 - 9~ 1 0 - 7mol L浓度的AngⅡ可使血管平滑肌细胞数明显高于对照组 (P <0 0 5 ) ;而用一定剂量超声辐照后 ,AngⅡ诱导的上述作用被明显抑制 (P <0 0 5 ) 。 To investigate the effects of ultrasound irradiation on proliferation of vascular smooth muscle cell in calves and to find out the best irradiation dose to inhibit the proliferation. Cultured calf aortic smooth muscle cells(CASMC)were divided into 4 groups. The first was control group;the second was exposed to ultrasound; the third was given angiotensin Ⅱ;the fourth was intervened by angiotensin Ⅱ and ultrasound irradiation. Cell counting, MTT and 3H TdR incorporation were used to determine the inhibition rate. Ultrasound irradiation for 60s at 2.2MHz,0.5 W/cm 2 inhibited the proliferation of VSMCs and the rate was 20%~30%( P <0.01).As the intensity increased, the time prolongd, the rate increased too. Ultrasound irradiation for 60s at 2.2MHz, 1 W/cm 2 could kill them. Ultrasound irradiation at a certain frequency and dose can inhibit the CASMC proliferation induced by AngⅡ.
