睡眠剥夺促进大鼠海马神经元凋亡及相关基因表达 被引量:7

摘要 探讨睡眠剥夺引起的神经元凋亡与相关基因表达的变化。采用TUNEL染色观察了快眼动睡眠剥夺大鼠海马神经元形态学变化 ,应用原位杂交、Westernblot法检测了快眼动睡眠剥夺大鼠海马bcl 2 ,baxmRNA ,MAPKs表达的变化。结果表明 :快眼动睡眠剥夺大鼠海马CA1 ,CA3区神经元阳性凋亡细胞数明显增多 ,bcl 2 ,baxmRNA表达明显增强 ,ERK活性降低 ,JNK蛋白表达量较对照组明显增高。提示睡眠剥夺可引起大鼠海马神经元凋亡。与凋亡相关的bcl 2 。 In order to explore neuronal apoptosis induced by sleep deprivation and the expression of some related gene. The morphology change of hippocampus neuron after paradoxical sleep derivation was observed by TUNEL method, the activity of MAPKs was assayed by Western blot and β liquid scintillation counting. The expressing changes of bcl 2 and bax mRNA were examined by in situ hybridization. The number of apoptosis of neuron in hippocampus increased, especially in CA1 and CA3 regions, ERK activity obviously lowered, and JNK expression increased followed by paradoxical deprivation. The results proved that the sleep deprivation could cause hippocampus neurons apoptosis and expressions of MAPKs and some genes which might be related to the mechanism of hippocampus neuron apoptosis.
