大鼠肺鳞癌中DFF45和Bcl-2基因表达的研究 被引量:1

摘要 目的:探讨DNA裂解因子(DFF45)和凋亡抑制基因bcl-2在大鼠肺鳞癌癌变转化过程中的表达及其关系。方法:Wistar大鼠60只,其中50只用化学致癌物3-甲基胆葸(MCA)及二乙基亚硝胺(DEN)碘油溶液于左肺叶支气管灌注诱发大鼠肺鳞状细胞癌,10只作为对照。应用免疫组织化学S-P法检测DFF45和bcl-2蛋白在癌变过程中的表达。结果:对照组大鼠支气管粘膜上皮细胞区、癌前病变区与肺鳞癌区中,DFF45蛋白的阳性表达率逐渐升高,分别为20.00%(2/10)、43.75%(7/16)、58.82%(20134),肺鳞癌区与对照组大鼠支气管粘膜上皮细胞区相比,DFF45蛋白阳性表达率的差异有显著性(P=0.0309):bcl-2蛋白的阳性表达率分别为:0、31.25%(5116)、47.06%(16/34),对照组大鼠支气管粘膜上皮细胞区分别与癌前病变区、肺鳞癌区比较差异均有显著性(P1=0.0495;P2=0.0065)。在大鼠肺鳞癌中,DFF45与bcl-2呈显著正相关(Pearson列联系数:0.5872,p=4.07×10^-6)。结论:bcl-2基因的过表达可能抑制了DFF45的裂解,从而使肿瘤细胞凋亡减少,增殖过多,协同促进了大鼠肺鳞癌的发生发展。
