摘要
用纯化的早孕因子(EPF)免疫BALB/C小鼠,取其脾脏与SP2/0小鼠骨髓瘤细胞融合,用酶联免疫吸附试验(ELISA)间接法筛选,以有限稀释法进行克隆化。杂交瘤细胞经过2~3次克隆后。阳检率达lOO%,建立了1B_6、2F_(10)二株分泌EPF单克隆抗体(EPF—McAb)的杂交瘤细胞系。用ELISA检测其诱生腹水效价均达1:1.28×10~5,初步鉴定该McAb具有敏感性高、特异性强的特点。应用EPF—McAb—ELISA对孕血清中的早孕因子进行检测,结果表明阳性率达92.9%。
To produce the monoclone antibodies to the human early pregnancy factor (EPF), BALB/C mice were immunized with EPF. The spleens were removed and the splenic cells were fused with mouse myeloma cells SP 2/0. The hybrid cells were cloned out by limited dilution. The cells were cloned 3 to 4 times untill the positive rate of clonal antibody to EPF approached 100%. The hybrid cells were injected into BALB/C mice introperitoneally. The ascites of the mice were harvested. The titer of the EPF McAb in the ascities was 1: 1.28×10~5by ELISA.
