摘要
目的 :探讨人肝癌中表达的 3种septin蛋白 1 ,2和6自身之间和相互之间是否存在相互作用关系 ,从而探讨在肝癌中septin家族的作用方式 .方法 :通过构建septin家族基因的pCMV Myc ,pCMV HA表达载体 ,采用人肾 2 93细胞内蛋白的免疫共沉淀进行验证蛋白的相互作用 .结果 :Septin家族蛋白中确实存在着相互作用 ,不仅蛋白相互之间存在相互作用 ,且同一种蛋白质的分子之间也存在着较强的相互作用 .结论
AIM: To study the interactions of three human septin proteins, septin 1, 2 and 6, and to investigate the working model of the septins in the hepatic carcinoma. METHODS: Full length septin 1, 2 and 6 gene were cloned into the pCMV Myc and pCMV HA vector respectively and co immunoprecipitation experiment in human 293 was used to investigate the interactions of the three septin proteins. RESULTS: Interactions in the septin family were confirmed, including the interactions between septin 1 and septin 2 or septin 6, and the interactions within themselves. CONCLUSION: Septin proteins as a complex may play an important role in the carcinogenesis by regulating cell cycles.
