bFGF对大鼠骨髓基质细胞的诱导分化作用

摘要 目的 观察bFGF对大鼠骨髓基质细胞的诱导分化作用。方法 从大鼠骨髓中分离培养基质细胞并传至第 3代 ,用含 10ng ml碱性成纤维细胞生长因子 (bFGF)的诱导培养基诱导 ,然后观察细胞形态的变化 ,并采用免疫组织化学法检测诱导后2 4h、48h、72h、96h细胞神经丝蛋白 (NF 2 0 0 )和胶质纤维酸性蛋白 (GFAP)的表达。结果 诱导后 2 4h的部分细胞不仅在形态上表现为神经元样 ,而且呈NF 2 0 0阳性表达 ,GFAP阴性。 48h、72h、96hNF 2 0 0阳性细胞数与 2 4h间无显著性差异 (P >0 .0 5 )。结论 bFGF可以在体外诱导骨髓基质细胞向神经元样细胞分化。 Objective To investigate the inducing effects of basic fibroblast growth factor (bFGF) on the differentiation of rat bone marrow stromal cells (rBMSCs) in vitro. Methods rBMSCs from rats were isolated and cultured for 3 passages, and then were treated with inducing medium containing 10 ng/ml (bFGF), 10% FBS (fetal bovine serum), DMEM/F12, 5 μg/ml insulin, 20 nmol/L progesterone, 20 nM glucose, 100 IU/ml penicillin, and 100 μg/ml streptomycin for 24, 48, 72, and 96 h. The cells were stained immunocytochemically with anti neurofilament-200 (NF-200) and anti glial fibrillary acidic protein (GFAP) antibodies. The morphological changes of the cells were observed. Results At 24 h after induction, part of the rBMSCs became neuron-like cells and showed NF-200 positive. No GFAP expression was found in the neuron-like cells. Compared with that at 24 h after induction, there was no significant difference in the number of NF-200 positive cells at 48, 72, and 96 h (P>0.05). Conclusion bFGF can induce the differentiation of rBMSCs into neuron like-cells.
