摘要
本文在钩体L型培养基中,应用不同浓度的青霉素G对波摩那型钩体进行L型体外诱导研究和形态学观察。结果表明:青霉素G在0.16~20u/ml均可诱导出钩体L型,其中以0.8~4u/ml诱导效果较好。用暗视野显微镜、Giemsa染色和扫描电镜检查表明,钩体L型在形态上表现为球状体和丝状体。前者大小不等。一般直径为0.15~4μm,大的囊样体直径约35μm,内部密度不匀,含大小不等的球状体;间接免疫荧光法检查表明,这些大小不等的球状体呈特异性阳性染色。透射电镜下钩体L型细胞壁有缺损,透光度大,有的L型细胞见DNA纤丝,研究还表明,37℃和29℃两种温度条件下诱导钩体L型的结果差异无显著性(P>0.05)
This paper reports leptospiral L- forms induced by penicillin G in the medium for leptospiral L- forms. The effective range of concentration was found to be 0.16 ~ 20 units per ml of media. Spherical bodies and filamentous forms of the organisms were observed in direct wet smear, after Gicmsa staining, and under scanning electron microscopy. The spherical cells varied in size from 0.15 to 4 μm in diameter and large cyst- like spherical bodies were around 35 μm in diameter. These spherical and cyst - like bodies were positive in indirect immunofluorcscent staining. Electron microscopy revealed that Icptospirdl L- forms were deficient in cell walls in parental organisms. This study also showed that there was no significant distinction in growth of the leptospiral L- forms at 37 ℃ and 29 ℃ (P>0.05).
