性激素结合球蛋白mRNA在子宫肌瘤组织中表达的研究

摘要 目的 研究子宫肌瘤组织和正常平滑肌组织中性激素结合球蛋白 (SHBG)野生型和突变型mRNA的表达 ,探讨SHBG与子宫肌瘤发生的关系。方法 选取 30例因子宫肌瘤行全子宫切除术的子宫标本 ,通过RT PCR及Southernblot研究子宫肌瘤组织及正常子宫平滑肌组织中SHBGmRNA表达。结果 野生型性激素结合球蛋白 (SHBGw)在子宫肌瘤组织与正常平滑肌组织的表达水平无显著差异 (P >0 .0 5 ) ,而外显子 (exon)VII缺失型SHBG(SHBGv)在子宫肌瘤组织中的表达水平明显低于正常平滑肌组织 (P <0 .0 5 ) ,SHBG在子宫肌组织表达水平与月经周期无关 (P >0 .0 5 )。结论  2种类型的SHBG在子宫平滑肌组织及子宫肌瘤组织均有表达 ,SHBGw可能与子宫肌局部高浓度激素的产生无关 ,而SHBGv低水平表达与子宫肌瘤的发生相关。 Objective To investigate the expression of sex hormone-binding globulin (SHBG) mRNA and the relationship between SHBG and uterine leiomyoma.Methods 30 samples of uterines by hysteromyomectomy were selected in the study. The RT-PCR and Southern blot were used to detect the expression of SHBG mRNA in tissues of uterine leiomyoma and normal myometrium.Results There was no significant difference in SHBG wild-type (SHBGw) mRNA between the tissues of uterine leiomyoma and normal myometrium(P>0.05), however, the SHBG exon VII splicing variant (SHBGv) levels in uterine leiomyoma was significantly reduced,as compared with those in normal myometrium (P<0.05).There was no obvious relationship between the levels of SHBG mRNA and the menstrual phase (P>0.05).Conclusion The expressions of both SHBGv and SHBGw can be found in both the tissues of uterine leiomyoma and normal myometrium.SHBGw may not be related to the high concentration of hormone in local myometrium,however, the low levels of SHBGv may be ralated to the genesis of uterine leiomyoma.
