摘要
目的:证明复方壳多糖人工皮肤在体促进创面愈合能力,为该人工皮肤应用于临床尤其是覆盖感染创面提供实验依据。方法:健康雄性大鼠24只,每只背部做全层皮肤缺损创面2个,每个创面涂1×105克隆形成单位(colonyformingunit,CFU)金黄色葡萄球菌,建立金黄色葡萄球菌污染创面大鼠模型,随机数字法分成实验组、对照组和空白对照组,每组16个创面,将复方壳多糖人工皮肤和复方胶原凝胶人工皮肤分别覆盖于实验组和对照组创面上,空白对照组无覆盖,通过细菌学、病理、免疫组化检查,了解两组之间的差别。结果:实验组和对照组的一般情况好,空白对照组差,实验组创面感染轻、创面收缩好,24h痂下细菌计量实验组为(8.09±2.51)×104CFU/g,明显少于对照组(5.91±2.51)×105CFU/g和空白对照组(5.55±2.18)×107CFU/g,差异有显著性意义;创周组织病理切片观察,实验组炎性细胞浸润少,愈合最快最好,对照组有炎性细胞浸润,空白对照组可见大量炎性细胞和渗出、出血、坏死;肌动蛋白免疫组化表明,实验组增生的纤维母细胞染色强阳性,对照组散在阳性,空白对照组染色阴性。结论:复方壳多糖人工皮肤有抗菌、抗感染和促进创面愈合作用,其作用优于复方胶原凝胶人工皮肤,是理想的创面覆盖物。
AIM:To identify the antibacterial ability of the composite chitosan artificial skin to accelerate wound healing so as to provide an experimental evidence for its application in clinic,especially in the covering contaminated wound. METHODS:Twenty four healthy male SD rats were divided into trial group,control group and blank control group evenly by means of random number table,with 16 wounds in each group.Each rat had two wounds on the back,with 1×105 colony forming unit(CFU) per wound painted with staphylococcus aureus,and then the rat model of wound contaminated by staphylococcus aureus was established.The composite chitosan artificial skins and the composite collagen artificial skins were sutured to the skin around the wounds in the trial and control groups,but nothing in the blank control group.The differences between the two groups were compared by bacteriological and pathological assay as well as immunohistochemical staining. RESULTS:All the rats of the trial group and control group had common condition,while those of the blank control group were bad.Compared with the control group,the wounds in the trial group was infected more lightly and had better contractility.Twenty four hours later,the average amount of bacteria under the crust of the trial group was (8.09±2.51)×104 CFU/g,less than that of the control group[(5.91±2.51)×105 CFU/g]and the blank control group[(5.55±2.18)×107 CFU/g],so there were significant differences between the trial and the two control groups.By the observation of histopathological sections around the tissues of wound in the three groups,the wounds in the trial group have least inflammatory cells infiltrated and the best wound healing,while there were less inflammatory cells infiltrated in the control group,and most inflammatory cell infiltrated in the blank control group with a great number of exudation,bleeding and necrosis.Actin immunohistochemical staining revealed that the fibroblasts were strongly positive in the trial group,weak positive in the control group and negative in the blank control group. CONCLUSION:Composite chitosan artificial skin has better antibacterial,anti infective and accelerating wound healing abilities than composite collagen artificial skin in tissue engineering,so it is optimal to cover wounds.
