摘要
目的:从基因水平探讨肿瘤坏死因子受体(TNF-αR1)在增生性瘢痕组织发生、发展过程中的作用,为增生性瘢痕的基因治疗探索一条有效途径。方法:以正常瘢痕为对照,利用RT-PCR技术检测1年以上增生性瘢痕组织成纤维细胞中TNF-αR1mRNA的表达情况。结果:1年以上增生性瘢痕组织中TNF-αR1mRNA的表达稳定,与正常瘢痕比较含量明显下降。结论:增生性瘢痕的形成与TNF-αR1的表达低下有一定关系。用基因治疗的方法提高早期增生性瘢痕中TNF-α的含量或者提高靶细胞膜上TNF-αR1的数目与活性可能为增生性瘢痕的康复治疗的有效途径。
AIM:To understand the role of tumor necrosis factor receptor (TNF αR1) in the occurrence and development process of hypertrophic scars based on gene level and to explore an effective way of gene therapy to cure the hypertrophic scars. METHODS: TNF αR1 mRNA expression in the hypertrophic scars over one year was tested by a semiquantitative reverse polymerase chain reaction (RT PCR) method as compared with normal scars. RESULTS:TNF αR1 mRNA expression in the hypertrophic scars over one year was stable and the number of hypertrophic scars significantly decreased as compared with normal scars. CONCLUSION:The formation of hypertrophic scars has certain correlation with the down regulating expression of TNF αR1.Gene therapy is an effective way for the rehabilitation treatment of hypertrophic scars to up regulate the concentration of TNF αR1 in early hypertrophic scars or increase the number and activity of TNF αR1 on target cell membrane.
