人类大肠腺癌组织中凋亡相关基因的研究(英文)

摘要 背景:bcl-2,c-myc基因在不同肿瘤中的表达和意义不尽相同。目的:探讨凋亡相关基因bcl-2和c-myc在人类大肠腺癌中的表达及其与预后的关系。设计:非随机对照的实验研究。地点、材料和干预:实验在解放军第四军医大学西京医院病理科完成。72例人类大肠腺癌选自西京医院外科手术及活检标本,5例正常人类结肠组织取自第四军医大学病理学教研室尸检组织(排除有结肠疾病)。运用HE染色和免疫组织化学方法对72例人类大肠腺癌及5例正常人类结肠组织进行研究,计算凋亡指数及免疫组化阳性率。主要观察指标:①不同分化程度的大肠癌凋亡指数。②大肠腺癌中Bcl-2及c-Myc的表达。③bcl-2和c-myc的关系。结果:大肠腺癌的细胞凋亡随肿瘤分化的不同,其差异有显著性意义(t=16.84,P<0.01),肿瘤分化程度越高,凋亡指数值越大。bcl-2和c-Myc在大肠腺癌中均具有较高程度的表达,两者的阳性率分别为61.1%和66.7%。Bcl-2与大肠腺癌的分化程度有关(χ2=11.29,P<0.05),肿瘤分化越低,其表达也就越低。c-myc在不同分化程度大肠腺癌的表达差异无显著性意义。Bcl-2和c-Myc在大肠腺癌组织中的阳性反应呈显著正相关(r=0.40,P<0.05)。结论:大肠腺癌细胞凋亡与肿瘤的恶性程度有关,bcl-2与c-myc均与大肠腺癌的细胞凋亡的调控有关。 BACKGROUND:The expression pattern and importance of bcl 2 and c myc genes were different in different tumors. OBJECTIVE:To investigate the expression of two apoptosis related genes,bcl 2 and c myc,in the regulation of apoptosis in human large intestine adenocarcinoma and to evaluate their relationship with the prognosis. DESIGN:A non randomized controlled experimental study was conducted. SETTING,MATERIALS and INTERVENTIONS:The experiment was conducted in the Department of Pathology,Xijing Hospital, Fourth Military Medical University.Seventy two samples of human large intestine adenocarcinoma specimens and five normal colon specimens(excluding colon disease) as controls were studied by HE staining and SABC immunohistochemistry.Apoptosis index and immunohistochemical positive rate were calculated. MAIN OUTCOME MEASURES:Apoptosis index of large intestine adenocarcinoma in diverse differentiation;expression of Bcl 2 and c myc in large intestine adenocarcinoma;relationship between bcl 2 and c myc. RESULTS:Cell apoptosis in large intestine adenocarcinoma varied significantly with the differentiation of neoplasm.The higher differentiation,the higher apoptosis index(t=16.84,P< 0.01).Bcl 2 and c Myc were all highly expressed in large intestine adenocarcinoma,of which the positive rates were 61.1%and 66.7%,respectively.However,the Bcl 2 expression changed with the differentiation of neoplasm(χ2=11.29,P< 0.05),i.e.,it was expressed weaker in the moderately and poorly differentiated specimens but higher in the highly differentiated specimens,while c myc had no such correlation with the differentiation of neoplasm.There was significantly positive correlation between Bcl 2 and c Myc immunoreactivity(r=0.40,P< 0.05). CONCLUSION:Both bcl 2 and c myc genes are correlated with the regulation of apoptosis in large intestine adenocarcinoma.bcl 2 may block the mechanism by which c myc induces cellular apoptosis,but it may not affect proliferous action induced by c myc,suggesting that c myc's regulation mechanisms on the cellular apoptosis and proliferation might not be identical.
