复方茯苓甘草汤对低氧大鼠肺血管构型重建的影响 被引量:6

摘要 目的 :从肺血管构型重建角度研究复方茯苓甘草汤对低氧性肺动脉高压 (HPH)的防治作用。方法 :采用间断常压氧法制备慢性HPH大鼠模型 ,右心导管法测量平均肺动脉压 (mPAP) ,应用光镜、免疫组化及半自动图像分析系统研究缺氧 2 1天后大鼠肺小血管病理变化及复方茯苓甘草汤的防治作用。结果 :低氧 2 1天后大鼠mPAP、右心肥大指数、肺中小型动脉相对中膜厚度、相对中膜面积、肺小血管肌型和部分肌型动脉百分比均高于正常组 (P <0 .0 1或P <0 .0 5) ;非肌型动脉百分比低于正常组 (P <0 .0 5) ;复方茯苓甘草汤可有效逆转上述改变。结论 :复方茯苓甘草汤的应用有望成为治疗HPH的有效措施 ,其机理与逆转肺血管的构型重建有关。 AIM: Investigate the effect of Fufangfulingancao Decoction on pulmonary vascular structure remodeling during the progression of pulmonary hypertension induced by hypoxia in rats. METHODS: Thirty healthy Spraque-Dawley male rats of three months old weighted (260±20)g was randomly divided into control group ( n =10) , hypoxia group( n =10) and Fufangfulingancao Decoction treatment group( n =10). Established the rat model of chronic hypoxic pulmonary hypertension by exposing rats to normobaric hypoxic conditions for 3 weeks, and rats of control group were used as normoxic controls. Mean pulmonary arterial pressure (mPAP) was measured by a right cardiac catheterization. Right ventricular hypertrophy was expressed as the ratios of right ventricular wall to left ventricle plus septum. The pulmonary vascular structural changes were observed with optical microscope, immunohistochemistry and Weigert elastic fibers staining. RESULTS: Hypoxic rats developed remarkable pulmonary hypertension and increment in the right ventricular hypertrophy ( P <0.01) after 21days' hypoxia, and the number of muscular and partially muscular arteries enhanced significantly ( P <0.01), relative medial thickness and relative medial area of medical membrane increased also ( P <0.01). The number of non-muscular arteries decreased (P<0.05). Pretreatment with Fufangfulingancao Decoction could attenuate the changes mentioned above. CONCLUSION: Fufangfulingancao Decoction can efficiently restrain HPH. Its mechanism maybe partly relates to the promotion of the pulmonary vascular structural remodeling.
