摘要 目的 研究犬脑枪弹伤后神经细胞凋亡机制及bcl 2的变化规律。方法 建立犬颅脑枪弹伤模型 ,制备不同时期、不同部位脑组织切片。采用末端脱氧核苷酸转移酶介导的生物素脱氧尿嘧啶核苷酸缺口末端标记法 (TUNEL)检测神经细胞凋亡以及用免疫组织化学方法检测bcl 2的动态变化。结果 对照组犬脑偶见神经细胞凋亡 ,神经细胞中有微弱bcl 2的表达 ,枪弹伤后凋亡神经细胞及bcl 2于 2h开始增加 ,2 4h达高峰 ,4 8h开始下降。不同区域表达不完全一致。各观测点神经细胞与bcl 2表达呈负相关。结论 犬脑枪弹伤后神经细胞bcl 2的表达增强 ,在不同时期、不同部位具有时空规律性。 Objective To investigate the mechanism of neuron cell apoptosis and the regulation of bcl-2 in cerebral cortex following craniocerebral gunshot wounds in canines.Methods An experimental craniocerebral gunshot wound model of canine was set up. The brain samples were taken at different time and areas. Canine neuron apoptosis cells were detected by terminal deoxynucleotidyl transferease-mediated biotinylated deoxyuridine triphosphate nick and labeling(TUNEL), and bcl-2 positive neurons were observed by immunohistochemical method. Results Feeble neuron apoptosis and bcl-2 were detected in the control group. Apoptosis and bcl-2 positive expression in neurons after craniocerebral gunshot wounds were increased at 2 h, reached the peak at 24 h, and began to decrease at 48 h.The expressions in different areas were not the same. A negative correlation was seen between bcl-2 expression and neuronal apoptosis. Conclusion Apoptosis and bcl-2 positive expression in neurons was increased after craniocerebral gunshot wounds in canines, and was detected to have the temporal and spatial regulation at different time and areas. The neuron cell apoptosis is related to the control of anti-apoptosis gene in neuron.
