摘要
目的 探讨单核细胞趋化蛋白 (MCP- 1/ JE)在重症急性胰腺炎 (severe acute pancreatitis,SAP)发病机制中的作用。方法 以 5 %牛磺胆酸钠逆行胆胰管注射制作大鼠 SAP模型 ,检测胰腺湿 /干重比和进行胰腺组织病理学评价 ,分别采用逆转录 -聚合酶链反应 (RT- PCR)和免疫组织化学方法 ,检测胰腺组织中 MCP- 1/ JE m RNA及蛋白的表达。结果 与假手术组相比 ,造模各组胰腺湿 /干重比在12 h内明显上升 (P<0 .0 1) ,假手术组无 MCP- 1/ JE蛋白的表达 ,SAP组胰腺腺泡细胞有阳性表达 ,胰腺组织内 MCP- 1/ JE m RNA的表达显著上调 (P<0 .0 5或 P<0 .0 1) ,并且与胰腺湿 /干重比及胰腺组织损害程度呈正相关。结论 MCP- 1/ JE在 SAP早期发病中可能起重要作用。
Objective To explore the potential role of MCP-1/JE in the pathogenesis of severe acute pancreatitis(SAP).Methods SAP was induced by retrograde infusion of 5% sodium taurocholate into the bili-pancreatic duct. Wet to dry weight ratio of pancreas was measured,and pathological changes of the pancreas were observed. Expression of MCP-1/JE gene and protein in rat pancreasis were evaluated by RT-PCR and immunohistochemistry. Results MCP-1/JE protein was detected in pancreatic acinus cells. Compared with that in controls,wet to dry weight ratio of pancreas increased greatly during 12 hours(P < 0.01). The expression of MCP-1/JE mRNA in the pancreas was significantly increased(P < 0.05 or P < 0.01) and was correlated with wet to dry weight ratio of pancreas and the pancreatic damage.Conclusions MCP-1/JE might play an important role in the pathogenesis of SAP.
