摘要
鉴于天然产物中活性成分含量低 ,结晶性组分易堵塞管道等问题 ,本文以 30 %穿心莲内酯浸膏原料为例 ,通过超临界CO2 萃取结晶技术来提高穿心莲内酯晶体的含量。结果发现 :在温度 5 5℃ ,时间 90min ,流量 15L/min时 ,当萃取结晶压力超过 2 0MPa后 ,穿心莲内酯的纯度超过 80 % ,并采用HPLC分析实验结果。新型超临界CO2 萃取结晶技术为开发高纯天然产物活性成分提供一条捷径。
The extraction and purification of natural products with less content is a challenge.In order to solve the problem,such as pipeline jam by crystal products,30% andrographolide was selected to purify by supercritical extraction and crystallization.The result showed that there was synchronous gradient crystallization of andrographolide in supercritical CO 2 extraction,moreover,the higher the pressure,the smaller and shorter the crystal shape.The content of andrographolide was raised from 30% to 82.3% under 20 MPa,at 55 ℃,within 90 min,ata flux rate 15 L/min.The supercritical fluid extraction and crystallization is a new technique for separation of natural products in high purity.
