应用AFLP技术对蔷薇进行遗传多样性研究的方法初探 被引量:7

摘要 五种不同蔷薇品种的基因组DNA经限制性内切酶酶切,连接特异性接头,用5条人工设计的与接头序列相识别的AFLP选择性引物,进行AFLP PCR扩增,经琼脂糖凝胶电泳检测扩增结果发现,不同蔷薇品种DNA的扩增结果具有明显差异。因此,AFLP技术是一种适宜于蔷薇的遗传检测方法。 Five strains R.davunica genomic DNA were digested with restriction enzyme.5 AFLP selective primers connected with specific adapter,which could identified adapter sequence,were used to carry AFLP-PCR.PCR products were identified on agarose gel electrophoresis.Some differences were found in AFLP of different stains of R.davunica.The results showed that AFLP technique was an effective method in genetic diversity monitoring of R.davunica.
