摘要
研究了草菇原生质体制备与再生的最佳条件。试验结果表明,溶壁酶浓度2%,温度32℃,pH自然,以0.8mol L甘露醇为渗透压稳定剂,用菌龄3d的菌丝酶解2h,可获得最佳原生质体再生效果。试验还分析了紫外线(UV)、60Co γ射线、硫酸二乙酯(DES)对草菇原生质体的诱变效应。
The optimal conditions of protoplast preparation and regeneration of Volvariella volvacea were studied. The experiment results showed that using mycelia cultured for 3d as testing material, 0.8mol/L mannitol as osmotic pressure stabilizer, enzymolysis with 2% lywallzyme for 2h at 32℃ under natural pH was the optimum conditions for V. volvacea protoplast preparation and regeneration.
