用高效液相色谱电化学检测法测定鸡蛋中多巴胺的残留量

Determination of Dopamine in Egg,Plasma and Internal Organs of Chicken by High-performance Liquid Chromatography with Electrochemical Detection

使用反相离子对高效液相色谱电化学检测法测定蛋鸡的蛋、血浆及组织中的多巴胺含量。样品在上柱前用过氯酸匀浆,离心后用酸性氧化铝吸附洗脱,然后用 Ultrosphere ODS柱(5 μm)分离。流动相由0.1 mol/L 磷酸二氢钠和0.24 mmol/L 三乙胺组成(内含0.36mmol/L 辛烷磺酸钠(OSA),0.1 mmol/L 乙二胺四乙酸二钠(EDTA)pH4.4)。对样品中多巴胺的分离获得满意的效果,灵敏度可达到100 pg,回收率为70%。应用本方法研究多巴胺饲料添加剂对鸡蛋质量的影响,为进一步应用该饲料添加剂提供了理论依据。

A method using reversed-phase ion-pair high-performance liquid chromatography with electrochemical detection for determination of dopamine in egg,plasma and tissues has been developed. The sample preparation required homogenization in perchloric acid and centrifugation before injection in- to the column.Ultrosphere ODS(5μm)column and a mobile phase consisted of a phosphate(0.1 mol/L NaH_2PO_4,0.24 mol/L triethyllamine,0.36 mmol/L octylsulfonate,and 0.1 mmol/L EDTA(pH4.4). The satisfactory effect of dopamine determination was achived.The detection sensitivity is 100pg.The presence recovery is 70%.The methed has been applied to study the effect of dopamine-feedadditive on egg qualities.We provide the theoretical evidence so as to widely apply the dopamine-feedadditive.
