细胞凋亡与BCL-2在葛根素干预新生鼠缺血缺氧性脑损伤模型中的表达 被引量:10

摘要 目的 探讨葛根素治疗新生大鼠缺氧缺血性脑损伤 (HIBD)模型的疗效。方法 将 2 8只 7日龄SD大鼠随机分为四组。其中葛根素治疗组、生理盐水对照组、HIBD模型组各 8只 ,均结扎右颈总动脉后吸入 8%O22h ,制成HIBD模型 ;假手术组 4只。治疗组用葛根素 1 g/kg体重腹腔注射 7d ,对照组用生理盐水腹腔注射 7d ,HIBD模型组为空白对照。 7d后处死 ,均采用原位末端标记 (TUNEL)检测细胞凋亡和SP染色BCL 2。结果 假手术组可见少量凋亡细胞 ;HIBD模型组与生理盐水对照组细胞凋亡数显著多于葛根素治疗组 ,P <0 .0 1 ;BCL 2的免疫强度低于葛根素治疗组 ,P <0 .0 5。结论 葛根素对细胞凋亡有抑制作用 ,对BCL 2蛋白的免疫强度有增强作用 。 Objective To investigate the therapeutic effect of purerarin on newborn rat hypoxic ischemic brain damage(HIBD) model.Methods 28 newborn rat of 7 days ole were randomized into the following four groups,the puerarin group,the normal saline control group,the HIBD control group and the pseudo operation control group.While the pseudo operation control group was composed of 4,each of the other three groups had 8 rats that had right carotid artery been ligated and were kept in hypoxic (8% oxygen) enviroment for 2 hours to produce the HIBD model.The puerarin group was given intraperitoneal injection of 1g/kg puerarin.The normal saline control group was intraperitoneally injected with the same dose of NS.The HIBD control group and pseudo operation group did not receive any intervention.Apoptosis was tested by TUNEL.Results Few apoptosis could be seen in the pseudo operation group.It was more prevalent in the HIBD control group and NS control group than that of the puerarin group( P <0.01).The BCL 2 immunity of the puerarin group was more intensive ( P <0.05).Conclusions There is apoptosis in the HIBD.Puerarin is not only an inhibitor of apoptosis,but also a good treatment in HIBD of newborn rats.
