系统性红斑狼疮患者血清抗核小体抗体和脱氧核糖核酸酶Ⅰ活性的研究 被引量:3

摘要 目的探讨血清抗核小体抗体和血清脱氧核糖核酸酶Ⅰ(deoxyribonucleaseⅠ,DNaseⅠ)的活性与系统性红斑狼疮(SLE)病情的关系。方法采用间接ELISA法检测SLE患者血清抗核小体抗体浓度,DNA-甲基绿比色法测定其血清DNaseⅠ的活性。结果SLE患者血清抗核小体抗体浓度明显高于正常对照,且与SLE病情活动程度、肾脏损害、血管炎及补体下降相关;SLE患者组血清DNaseⅠ活性低于正常对照组,且与病情活动程度相关;血清抗核小体抗体浓度与DNaseⅠ的活性成负相关。结论抗核小体抗体是SLE的主要自身抗体之一,核小体刺激机体产生抗体的可能原因是DNaseⅠ的活性下降。 Objective: To study the correlations among the serum antinucleosome antibodies, DNase I activity and SLE activity. Methods: The serum antinucleosome antibodies and DNase I activity were detected respectively by indirect ELISA and DNA-methyl green substrate colorimetry, respectively. Results: The serum antinucleosome antibodies were significantly higher in SLE patients than those in healthy control. The increasing were associated with the disease activity, nephritis, vasculitis, and the decreased complement. The serum DNase I activity were significantly lower in SLE patients than those in healthy control, which correlate with the disease activity. There was an inverse correlation between the DNase I activities and anti-nucleosome antibodies in SLE patients. Conclusions: The anti-nucleosome antibody is one of the major autoantibodies against SLE and the decreased DNase I activity is one of the causes to production of antibodies against nucleosome antigens.
