摘要
采用5株小鼠肝炎病毒(MHv1、MHV3、A59、JHM和沪一79)研制了多价抗原检测大鼠冠状病毒(RCV)抗体的FLISA试剂盒,各株抗原等量混合.多价抗原包被浓度以1O一12μg/ml为宜。经99%可信限测定45只清洁级大鼠血清,N值为0.134,阳性临界值为0.281。用MHV阳性血清对多价MHV抗原阻断抑制卒为大于50%,而对Sendai、PVM和Reo3抗原其抑制率分别为17%、26%和33%。用多价抗原检测自然感染大鼠血清中的RCV抗体较用单价抗原敏感。本试剂盒对155只普通级大鼠测得阳性卒为90%,对46只清洁级大鼠阳性率为0%。本文还证明SPA酶结合物不适用于大鼠的检测。
This paper reported that ELISA kit for detection of antiobody to rat coronavirus(RCV)was developed with polyvalent antigen of 5 stains of MHV(NHV1;MHV#;A59;JHM;Sh一79).Equivalent monovalent antigen ofMHV 5 strains with a protein con-centration of 10一12μg/ml showed the best result fot coating.All of 45 elear rats gave a normal OD value(0.134)based on the 99%ofconfidence limit .Cut off value was 0. 281, Inblocking ELISA with polyvalent antigen,more than50%inhibition can be obtained by tis-ing MHV positivc scra in ELISA whereas,the percentage of inhibition were 17%, 28%and 33%in Sendai, PVM and Reo3 blockingELISA,respectively.Compared with monovalent antigen.polyvalent antigen showed a higher sensitivity for detection of natural infectionof RCV. Tghe results of serologieal monitoring using ELISAkit revealed that the rate of antibody-positive was 90%and 0%in conven-tional and clear ratrespectively.The results also demonstrated that the conjugate( SPA-HRP)was unsuitable to serologieal survey for rats.
