rhIL-3和rhGM-CSF对人正常骨髓造血干细胞体外维持的作用

摘要 应用重组人白细胞介素3(rhIL-3)和重组人粒-巨噬细胞集落刺激因子(rhGM-CSF)对18例正常人骨髓造血干细胞体外悬浮培养1周后,用琼脂集落培养测定CFU-CM的增殖程度。rhIL-3组的CFU-GM增至4.9±1.9倍;rhGM-CSF组增至4.7±1.6倍;rhIL-3+GM-CSF组增至11.5±5.4倍,均高于对照组(P<0.001)。rhIL-3+GM-CSF组的CFU-GM增殖量明显高于rhlL-3组和rhGM-CSF组。结果表明:rhIL-3和rhGM-CSF均能促使造血干细胞在体外短期维持中大量增殖,二者联合应用有协同作用。 The effects of rhIL-3 and rhGM-CSF on normal human bone marrow hematopoietic progenitorcells were studied in vitro. Culture treated with rhIL-3 induced an increase of CFU-GM up to 4.1+1.9 times; Culture treated with rhGM-CSF induced an increase of CFU-GM up to 4.7+1.6 times;Culture treated with rhIL-3+GM-CSF induced a significant increase of CFU-GM up to 11.3+3.4times. It was showed that both rhIL-3 and rhGM-CSF were the effective stimulating factors, whichcould enhance proliferation of hematopoietic progenitor cells in vitro. They were useful in maintainingand amplifying hematopoietic progenitor cells in vitro. They exhibited sinergistic activities when thesetwo factors were added together.
