胰岛素与环核苷酸衍生物对β射线转化细胞DNA合成的影响

摘要 本实验研究胰岛素与环核苷酸衍生物对鼠胚成纤维细胞(C_3H_(10)T1/2CL8,简称NC_4—H_(10))及其β射线恶性转化细胞(简称TC_3H_(10))DNA合成的影响。结果表明,胰岛素对两种细胞的DNA合成都有促进作用,对TC_3H_(10)的作用远大于NC_3H_(10)。双丁酰环—磷酸腺苷对两种细胞的DNA合成有抑制作用,双丁酰环—磷酸鸟苷对两种细胞的DNA合成有刺激作用。胰岛素与两种药物无协同作用。 We have investigated the regulation of DNA synthesis in C3H mouse embryo cells (C3H10T1/2CL8, NC3H10) and ~3H-TdR transformed mouse embryo cells (TC3H10) by insulin. Both cells werecultured in DMEM medium and assessed by ~3H-TdR incorporation. insulin stimulated DNA synthesisin both NC3H10 and TC3H10 cells, and had much more effect on TC3H10 cells than NC3H10 cells.0.05μg/ml insulin increased DNA synthesis by 39% in NC3H10 cells and by 103% in TC3H10 cells.The results also showed that generally db-cAMP had inhibition effect and db-cGMP had stimulationeffect on both cells. While adding insulin to db-cAMP or db-cGMP, the regulation effects were theadditive effects of either one alone.
