淋巴因子活化杀伤细胞抗恶性肿瘤的研究

摘要 由人外周血分离单个核细胞,在体外与白细胞介素2、植物血球凝集素及辅助细胞共同培养4周,获得大量活化的LAK细胞。表型及体内外生物学实验结果表明,LAK细胞为CD_3^+淋巴细胞,伴不同比例的CD_4^+与CD_8^+亚群,其抗肿瘤功能表现在体外对多种肿瘤靶细胞的溶解作用,在裸小鼠体内则能有效地抑制高转移癌系的成瘤及实验性转移。 Mononuclear cells (MNC) isolated from peripheral blood were co-cultured in vitro with recombi-nant human interleukin-2, phytomagglutinin (PHA) and assistant cells. The MNCs were activated intoLAK cells during the 4-week long term expansion. Phenotypic analysis showed that the LAK cells weremainly CD^+_8 lymphocytes with different ratio of CD^+_4 and CD^+_8 subpopulation after three weeks ofculture. Through ^(51)Cr-releasing assay, it had been proved that LAK cells would lyse several kinds oftumor targets, including fresh and cultured tumor cells, with no effect on normal cells. It was alsoconfirmed that LAK cells could inhibit the growth and metastasis of highly metastatic tumor lines (PGtumor) during the Winn's tests and experimental metastasis study in nude mice.
