摘要
以SO-Rb50第327代细胞,提取其基因组DNA,以正常人血细胞为对照,检测其Rb抗癌基因和c-myc癌基因。结果表明,该细胞株Rb基因完全缺失,c-myc原癌基因扩增3倍。此为以该细胞株在体外进一步研究肿瘤生长调控和逆转提供依据。
Retinoblastoma (Rb) is the most common malignant cancer of the eye. SO-Rb50 is the first Rbcell line established in China in 1988. It has passed to the 387th generation now. we collected cells ofthe 327th generation of SO-Rb50, Purified their genomic DNA and detected them with Rb and c-myccDNA probes respectively (normal human white blood cells DNA was the control). We found theRb gene was deleted while c-myc gene was three fold amplified, Which provides a basis for furtherstudy of the regulation of tumor development and tumor reversal with this cell line in vitro.
