神经胶质瘤中受体酪氨酸激酶信号传导系统及酪氨酸激酶抑制剂的研究进展

摘要 原发性的神经胶质瘤 ,特别是多形性胶质母细胞瘤 ,其生物学特征如不可控制的细胞增生 ,正常凋亡程序的关闭 ,新生血管的形成和浸润性生长 ,给治疗造成了极大的困难。目前认为一系列的基因变异是发生胶质瘤的生物学基础 ,这些变异主要发生在受体酪氨酸激酶信号传导系统。该系统可调控细胞周期 ,调节细胞生长与分化 ,促进损伤修复。最近的研究表明 ,受体酪氨酸激酶信号传导系统的异常活化与胶质瘤的发生和进展密切相关。酪氨酸激酶抑制剂可选择性抑制酪氨酸激酶的活性 ,从而阻断肿瘤细胞的信号传递 ,抑制肿瘤的生长 ,增加肿瘤对放、化疗的敏感性。对胶质瘤中受体酪氨酸激酶信号传导通路及酪氨酸激酶抑制剂的研究 ,是当今国内外生物治疗肿瘤的研究热点之一。
