非小细胞肺癌DNA含量与CD44和Fas表达的相关性研究 被引量:1

摘要 目的 观察转移相关基因CD44和Fas蛋白在肺癌中的表达及与DNA含量的关系。方法 应用流式细胞术对 40例肺癌患者手术新鲜标本中CD44和Fas蛋白表达及DNA含量进行检则 ,并与正常对照组 (n =2 5 )和肺部良性病变组 (n =2 0 )进行对比研究。结果 肺癌组Fas表达低于正常对照组和肺部良性组 ,CD44表达和DI值高于正常对照组和肺部良性组 ,正常对照组中CD44和Fas表达及DI值与良性病变组相比差异均无显著性。Ⅰ +Ⅱ期肺癌组织中Fas的表达高于Ⅲ +Ⅳ期 ,Ⅰ +Ⅱ期CD44表达水平和DI值则低于Ⅲ +Ⅳ期 ,中高分化与低分化之间CD44、Fas表达和DI值差异有显著性 ,肺癌伴淋巴结转移者其CD44表达高于不伴淋巴结转移者 ,Fas表达和DI值与有无淋巴结转移无明显的相关性。随着肺癌细胞DI值不断升高 ,Fas表达逐渐降低 ,与DI值呈负相关 (r =-0 44 3 ,P <0 0 1) ;CD44表达则逐渐升高 ,呈正相关 (r =0 64 6,P <0 0 1)。在CD44、Fas ;表达及DI值与肺癌临床参数的研究中 ,发现CD44、Fas表达和DI值与患者的年龄、性别、肿瘤大小和病理类型均无明显的相关性。结论CD44、Fas表达及DNA含量在肺癌发生、发展和转移过程中起着共同调节作用 。 Objective To observe the relationship between expression of CD44、Fas and DNA content in lung carcinomas. Methods The expression of CD44、Fas and DNA content in 40 patients with lung cancer were detected by flow-cytometry,normal control groups (n=25)and 20 patients with benign lesions served as contrtols .Results The expression of Fas protein in lung cancer were lower than those in normal control groups and benign lesion groups, The expression of CD44 protein and DNA index (DI)in lung cancer were higher than those in normal control groups and benign lesion groups,There was no significantly difference between the expression of CD44 and Fas and DI in normal control groups and benign lesion groups. The expression of Fas in stage Ⅰ+Ⅱ were higher than those in stage Ⅲ~Ⅳ,the expression of CD44 and DI were lower than those in stage Ⅲ+Ⅳ.There was a distinct statistic difference between CD44、Fas and DI in moderate-well histopathological grading and those in poor histopathological grading. The expression of Fas Was no related to his topathological grading.The expression of CD44 were closely correlated with lymph node metastasis,primary lung carcinoma with lymph node metastasis showed no higher Fas and DI than lung carcinoma without lymph node metastasis. The DI gradually increased with tumor progression, theexpression of Fas protein were gradually decreased, There was an inversely correlation between the expression of Fas and DI(r=-0 443,p<0 01);the expression of CD44 was also closely increased, there was an positively correlation between the expression of CD44 and DI(r=0 646,p<0 01);these parameters were not correlated with sex、 age 、tumor size and pathological type.Conclusion CD44 and Fas and DNA content probably play an important regulating role in the genesis、development and metastasis of lung cancer and they may be as a index to forecast potential of the rumor metastasis.
