摘要
目的:探讨肥大细胞对眼眶成纤维细胞生长的影响。方法:将分离纯化的小鼠骨髓源性肥大细胞(BMMC)与眼眶成纤维细胞(OFb)共育,共育分为两组:两种细胞接触共育组和非接触共育组,眼眶成纤维细胞单独培养为对照组;应用光镜、电镜及细胞生长计数等方法研究肥大细胞对眼眶成纤维细胞生长的影响。结果:与BMMC接触共育的OFb增殖活跃,呈复层生长,单位面积内细胞计数明显增加;接触共育组中OFb的增殖数分别与非接触共育组和对照组比较,差异均有统计学意义(P<0.01);而非接触共育组的OFb增殖数和对照组比较,差异没有统计学意义(P>0.05);电镜见接触共育组OFb中粗面内质网(RER)增生扩张,BMMC与OFb紧密接触,部分BMMC有脱颗粒现象。结论:肥大细胞对眼眶成纤维细胞的生存和增殖有显著的促进作用,并使其功能活跃,这种作用主要是通过两种细胞间的紧密接触而实现的。
Objective:To explore the effects of mast cell on the proli feration of orbital fibroblasts (OFb). Methods:To separate and purify bone marr ow-derived mast cells (BMMC) and orbital fibroblasts (OFb) of mice. Three groups were designed in the study: the group of OFb cultured solely (as control), the group of BMMC and OFb cocultured contactly(test group) and the group of BMMC and OFB cocultured uncontactly. Light microscope and electron microscope and cell c ount technique were used to investigate the effects of mast cells on the prolife ration of OFb. Results:The proliferation of OFb in test group was very active, a nd presented multilayer growth tendency, and the number of the OFb significantly increased comparing with that of group three and the control group respectively (P <0.01). Whereas the number of the OFb in group three had no significant dif ference comparing with the control (P >0.05). Under electron microscope, the RER increased obviously and BMMC closely contacted with OFb in group two, and some BMMC presented degranule phenomena. Conclusions:Mast cell can obviously promote the survival and proliferation of orbital fibroblasts, and active the functions of OFb. The effects of mast cell on OFb may be completed by tight contaction bet ween BMMC and OFb.
