摘要
目的:观察脑缺血后不同时间段IL-6在脑缺血区和血清的表达及电针对其表达的调节。方法:通过大脑中动脉线栓法建立SD大鼠局灶性脑缺血模型,于造模成功后电针相应组大鼠,再利用免疫组化SP法、放射免疫分析技术,检测脑缺血及电针对IL-6表达的影响。结果:脑缺血后IL-6免疫阳性细胞在脑缺血区表达增加,3d达高峰,平均细胞数为(28.15±6.08)个/0.375mm2;血清IL-6水平也升高,1d达高峰,浓度为(103.79±13.65)pg/ml。电针可明显提高IL-6的表达。缺血+电针组与缺血组比较,缺血+电针1、3d组的IL-6阳性细胞数增加(P<0.05),缺血+电针3、7d组的血清IL-6水平升高(P<0.05)。结论:电针上调IL-6的表达,可能是电针治疗缺血性脑损伤的机制之一。
Objective: To investigate the expression of IL-6 in the cerebr al ischemic area and serum and explore the effects of electroacupuncture(EA) on the expression at various time points after establishing a model of cerebral isc hemia in rat. Methods: In order to prepare focal cerebral ischemia model, the mi ddle cerebral artery (MCA) of Sprague-Dawley rat was occluded by a monofilament. Later, rats in ischemia+EA group were subject to EA. Immunohistochemical techni que and radioimmunoassay were used to detect the influence of cerebral ischemia and EA on IL-6 expression of rats. Results: The number of IL-6 positive cells in the cerebral ischemic area increased after ischemia and reached the peak about 28.15± 6.08 cells/0.375mm . Serum IL-6 level also increased with a peak of 103 .79±13.65 pg/ml at 1 day after ischemia. EA could raise the expression of IL-6 both in the cerebral ischemic area and serum. As compared with ischemia group, the number of IL-6 positive cells of ischemia+EA 1?3d group was raised(P <0.05)a s well as serum IL-6 level of ischemia+EA 3?7d group. Conclusions: The up-regula tion of IL-6 may be one of the mechanisms of EA protecting against ischemic cere bral damage.
