反相制备色谱法分离葛根素

摘要 葛根采用溶剂冷浸提取3次,每次48h,提取物经酸水解获得粗品葛根素。粗品葛根素经反相制备色谱分离纯化,分离条件为:YWG-C18反相色谱柱(22×300mm,30μm),流动相为甲醇,流速为2.0mL/min,紫外检测,波长为270nm,所得产品经薄层层析鉴定纯度。 The roots of Pueraria lobata (Willd) Ohwi were soaked with methanol for two days(48 h). The extract was hydrolyzed with 5% HCl to give crude puerarin. The reversed phase liquid chromatography (RP-LC) method was used to separate puerarin and the separation was performed on YWG-C18(22×300 mm,30 μm) column, eluted with methanol. The wavelength for detected was 270 nm. Then colorless needle crystal was obtained by recrystaling with glacial acetic acid-methanol that their volume ratio is 1∶1. Purified puerarin was identified by TLC.
