参附注射液对离体大鼠缺氧/复氧心肌的保护作用 被引量:19

摘要 目的 探讨参附注射液对缺血 /再灌注损伤心肌的保护作用及其机制。方法 采用Langendorlf大鼠离体心脏灌流技术 ,制备心肌缺氧 /复氧损伤模型 ,分别在缺氧前及缺氧后 ,用参附注射液对心肌给予保护 ,利用高效液相色谱仪(HPLC)测定离体大鼠心肌组织中的高能磷酸化合物的含量变化 ,利用H— 6 0 0透射电子显微镜对心肌细胞超微结构进行观察。本实验共分 4组 :正常对照组 ,单纯缺氧 /复氧组 ,参附注射液前、后保护组。结果 ①单纯缺氧 /复氧组心肌组织中AMP、ADP、ATP及AN含量均明显低于正常对照组 (P <0 .0 1)。②参附注射液前、后保护组大鼠心肌组织内的AMP、ADP、ATP、AN含量均高于单纯缺氧 /复氧组 (P <0 .0 1) ,且接近正常水平 (P >0 .0 5 )。电镜结果也证明参附注射液对缺氧 /复氧心肌细胞超微结构具有明显的保护作用。结论 参附注射液无论在缺氧前预灌注时及缺氧后再灌注时给予 ,均能通过提高心肌组织中高能磷酸化合物的含量 。 Objective The experiment was designed to study the effect of compound Shenfu injection (SF) on myocardium with anoxia/reoxygenation. Methods The myocardial anoxia/reoxygenation model was made in perfused isolated rat heart. SF was given at the time of pre - perfusion and reperfusion,then HPLC and H - 600 electron microscope were used to detect the change of high energy phosphate and the ultrastructure of myocardial cell. Results ①The contents of AMP, ADP, ATP and AN in myocardium in only anoxia/reoxygenation group were obviously lower than those in the control group (P<0.01 ). ②The contents of AMP, ADP, ATP and AN in myocardium in the groups with SF were higher than those in only anoxia/reoxygenation group (P<0.01 ). In those groups with SF, the contents of ATP and AN were close to normal level (P>0.05). Conclusions These results indicated that SF administration could significantly increase the content of high energy phosphate in myocardium with anoxia/reoxygenation and decrease the ultrastructure injury of myocardial cells.
