摘要 [目的]研究大豆异黄酮诱导裸鼠EC鄄7906移植瘤凋亡的机制。[方法]建立人食管癌EC鄄7906裸鼠移植瘤模型,采用不同剂量的大豆异黄酮进行瘤旁注射,并设生理盐水和二甲亚砜(DMSO)两个对照组,用透射电镜和TUNEL法检测肿瘤组织细胞凋亡情况,免疫组织化学法和RT鄄PCR法检测肿瘤组织的凋亡相关基因bcl鄄2和bax的表达情况。[结果]透射电镜和TUNEL法发现大豆异黄酮导致移植瘤内大量细胞发生凋亡;免疫组织化学法发现大豆异黄酮注射组瘤组织的bcl鄄2蛋白表达阳性率降低,bax蛋白表达阳性率明显提高;RT鄄PCR法发现大豆异黄酮注射组的bcl鄄2mRNA条带密度随剂量的增大而递减,baxmRNA条带密度递增。[结论]大豆异黄酮对人食管癌EC鄄7906裸鼠移植瘤具有抑制作用,通过下调bcl鄄2的表达和上调bax的表达而诱导食管癌移植瘤细胞发生凋亡。 To investigate the molecular mechanism of the apoptosis induced by soybean isoflavone in transplanted tumor of human esophageal cancer cells EC-7906 in nude mice.Human esophageal cancer cells EC-7906 were implanted into nude mice to get the cancer model of EC-7906.Soybean isoflavone with different doses was injected around the tumor on the nude mice.After treatment, transmission electron microscope and TUNEL staining method were used to detect the apoptosis status of transplanted tumor cells. Immunohistochemical staining and RT-PCR were used to detect the expression of apoptosis-associated gene bcl-2 and bax in transplanted tumor.Apoptosis in numerous cells of tranplanted tumor induced by soybean isoflavone was detected by transmission electron microscope and TUNEL staining method.Low-expression of bcl-2 gene and high-expression of bax gene were detected by immunohistochemical staining and RT-PCR.[Conclusions]Soybean isoflavone has inhibiting effect to transplanted tumor of human esophageal cancer cells EC-7906 in nude mice.Soybean isoflavone can induce apoptosis in transplanted tumor of esophageal cancer cells EC-7906.This apoptosis may be induced by up-regulating the expression of bcl-2 gene and down-regulating the expression of bax gene.
