摘要
目的 研究外源性内皮素 - 1(ET - 1)对经 5 -氮胞苷体外诱导兔骨髓间质干细胞分化的心肌样细胞转录因子GATA - 4磷酸化的影响。方法 取幼年兔股骨干骨髓基质细胞经 5 -氮胞苷诱导分化为心肌样细胞 ,分别加入不同浓度 (10、30、5 0nmol/L)的外源性ET - 1,Western -blot分析心肌细胞GATA - 4转录因子的蛋白表达及其磷酸化的改变。结果 分化的心肌样细胞可表达心肌细胞特异性GATA - 4转录因子 ;外源性ET - 1不影响GATA - 4蛋白表达 ,但可快速诱导GATA - 4转录因子的磷酸化 ;其中 ,30nmol/LET - 1时GATA - 4转录因子磷酸化的程度最高。结论 ET - 1可介导 5 -氮胞苷诱导兔骨髓间质干细胞分化的心肌样细胞GATA - 4转录因子的磷酸化而影响其活性。
Objective To investigate the influence of exogenous endothilin-1 (ET-1) on phosphorylation of cardiac transcription factor GATA-4 in cardiac-like cells induced from rabbit bone mesenchymal stem cells (BMSCs). Methods The BMSCs from the shaft of femur of young rabbit were cultured in vitro and induced into cardiac-like cells with 5-azacytidine. Different concentrations of ET-1 were added to induced cardiac-like cells(10 nmol/L, 30 nmol/L and 50 nmol/L). Western blot was employed to analyze the alterations of protein expression and phosphorylation level of GATA-4. Results The expression of specific GATA-4 was noticed in differentiated cardiac-like cells. Exogenous ET-1 could induce phosphorylation of GATA-4 quickly, but did not affect GATA-4 protein expression. When 30 nmol/L ET-1 was added, the phosphorylation of GATA-4 reached its highest level. Conclusion Exogenous ET-1 can induce phosphorylation of cardiac transcription factor GATA-4 and influence the activity of GATA-4.
